Opportunity
Zinc pyrithione (ZPT) is a widely used anti-dandruff agent in shampoos and personal care products, as well as an effective bactericidal and fungicidal compound in industrial applications such as antifouling paints and plumbing systems. Despite its widespread use, ZPT poses significant risks to human health and the environment. Studies have shown that ZPT can penetrate human skin, disrupt zinc homeostasis in cells, and cause DNA damage, oxidative stress, and even reproductive toxicity. Ecotoxicological studies further reveal its persistence in aquatic environments, where it harms marine life, including fish larvae, polychaetes, and mussels. Regulatory bodies like the European Commission have proposed bans on ZPT in cosmetics due to these hazards. Current detection methods, such as high-performance liquid chromatography-tandem mass spectroscopy (HPLC-MS), are costly, time-consuming, and require complex lab equipment. Alternative techniques like UV/Vis spectroscopy or electrochemical sensing are prone to interference and lack sensitivity. There is an urgent need for a rapid, low-cost, and on-site detection method to monitor ZPT in complex matrices like shampoos, wastewater, and biological samples.
Technology
This patent introduces an innovative method for detecting ZPT using surface-enhanced Raman spectroscopy (SERS). The technology involves a substrate coated with metal nanostructures (e.g., silver or gold nanospheres), which amplifies Raman signals when exposed to ZPT-containing samples. The substrate is fabricated by electrochemically depositing metal ions (e.g., Ag, Au) onto a base material (e.g., acupuncture needles). When the substrate contacts the sample (via immersion or spraying), ZPT molecules bind to the nanostructures, generating distinct Raman peaks at 575, 829, 1136, and 1545 cm⁻¹. These peaks correspond to ring deformation, ring breathing, C–H deformation, and ring stretching vibrations, respectively, providing a fingerprint for ZPT identification. The method achieves exceptional sensitivity (detection limit: 0.003 ng/mL) and linear quantification (0.03–8 µg/mL) without sample pretreatment. The substrate’s selectivity ensures ZPT detection even in complex mixtures (e.g., shampoos), as ZPT preferentially binds to the nanostructures over competing molecules like rhodamine 6G.
